Effect of Phytoestrogen on Nitric Oxide Synthase in Ovariectomized Rabbits

Abstract
|
|
|
Objectives: To elucidate the action of phytoestrogen on nitric oxide synthase in ovariectomized rabbits.
Methods: Twenty young New Zealand White Rabbits were studied for 19 weeks. The animals were divided into five groups: (I) 4 normal diet, (II) 4 ovariectomized (OVX) plus normal diet, (III) 4 OVX plus 0.5% (w/w) cholesterol diet, (IV) 4 OVX plus 0.5% cholesterol diet plus 15% (w/w) Supro plus 675¨Þ (soybean concentrate), and (V) 4 OVX plus Supro plus 675¨Þ group.
Results: The blood 17 estradiol-2 levels were decreased in the OVX groups (II, III and IV) without significance. The nitric oxide level was slightly increased only in the OVX plus Supro plus 675¨Þ group (V). The blood cholesterol was increased significantly in the two cholesterol-treated two groups (III, IV). The high density lipoprotein (HDL) was decreased in cholesterol-treated two groups significantly (III, IV). The low density lipoprotein (LDL) was only increased in the OVX plus cholesterol group (III). Endothelial NOS expression in thoracic aorta was more increased in the cholesterol and/or Supro plus 675¨Þ groups (III, IV and V) than in the I and II groups. Brain (nervous) NOS was decreased in the Supro plus 675¨Þ treated groups (IV and V).
Conclusion: These results suggest that phytoestrogen prevents LDL increase caused by hyperlipidemic diet. Phytoestrogen increases eNOS expression in aorta of ovariectomized rabbits, however, it inhibits bNOS production in the brain. Phytoestrogen seems to have an estrogen-mimic action on blood vessels. Inhibited bNOS in the brain may be related to the beneficial action of phytoestrogen on the brain.
